Glance doesn’t seem to care if an instance that has been launched from the image is running. See the API documentation.I tried this on Newton and was able to remove the omage without complaint.

I can’t imagine that instances access Glance when they are restarted. If the instance disk is a volume, they start from the volume. If it is ephemeral, they start from the local copy of the image on the compute host.
